If you are in the process of applying for a new job and you have got your CV up to date, all you need to do now is to do a covering letter to send with your CV. To do a good covering letter you need to write it in accordance with the job that you are applying for and go through the job advert that you have seen that has enticed you to apply for the job in the first place.
It is important to have both the advert and your CV with you when writing this letter so that you can tailor it to the letter and show your potential new employers that you have paid particular attention to this role and not just sent them a blanket cover letter.
If you would like an example cover letter to send with your CV, please see the below example which you can use as a guide:
Cover Letter for a Job
Dear Sir/Madam,
Application for the role of (NAME OF POSITION and ANY JOB REF)
I would like to submit my CV in application for the (NAME OF POSITION) which you have advertised in (what publication/online or other and what date).

I have excellent communication skills, I am very organised and I pay great attention to detail. I enjoy working with others and I am always keen to learn new skills.
I have enclosed my CV for your consideration. Should you have any questions about my skills and experiences please do not hesitate to contact me.
Thank you for taking the time to read my letter and CV and I look forward to hearing from you.
Yours sincerely

If you are applying for a job and doing an application form and looking to write a cover letter to go with your application then you need to write your letter based on the application form that you have already completed and focus on the role that you are applying for.
Most of what you want to say in a covering letter will already be in your application as forms tend to be very detailed so you may feel that you are covering it again but bear in mind that someone will be reading your letter before your application form so it is important to put the key and most important parts into your letter so if necessary they read it twice and remember it.
